Handling RNA

Isolating RNA

*For isolating RNA from cells and tissues, use fresh samples or samples that have been stored at or below -70°C.  This reduces RNAse activity.

*RNAse inhibitors can be used to protect RNA from degradation during isolation and in all later applications.  To keep the inhibitor active, avoid using strong denaturing agest like SDS or urea, and maintain reducing conditions (such as by using DTT) and hold the temperature below 65°C.

Storing RNA

*Store RNA aliquoted in ethanol or isopropanol at -80°C.

*Remove ethanol or isopropanol later by centrifugation and resuspend in the appropriate RNAse-free buffer.

Dissolving RNA

*Dissolve RNA in RNAse-free buffer or water and incubate on ice for 15 minutes.

*If it doesn't dissolve well, gently invert and/or tap the tube several times.
